Combination of single-molecule fluorescence and mass photometry

چکیده
Total internal reflection fluorescence microscopy can detect and image single fluorescently labeled molecules close to a surface with high temporal spatial resolution. Despite the specificity introduced by fluorescent label, it is not universal, label must be attached individual proteins through genetic engineering or designed antibodies. Mass photometry (MP) interferometrically detects elastic scattering rather than of particles landing at glass-water interface, enabling only detection, imaging localization, but also mass measurements proteins.

Common uorescent agents include organic uorophores (e.g., Cy5), uorescent proteins (e.g., GFP), and semiconductor quantum dots each with different size, photobleaching, blinking, and labeling ef ciency properties. Wideeld detection with EMCCD camera enables parallel measurements. The temporal resolution is >ms, and the working concentration is limited to nM or less. Confocal detection from an e...
متن کاملEnhancement and quenching of single-molecule fluorescence.
We present an experimental and theoretical study of the fluorescence rate of a single molecule as a function of its distance to a laser-irradiated gold nanoparticle. The local field enhancement leads to an increased excitation rate whereas nonradiative energy transfer to the particle leads to a decrease of the quantum yield (quenching). Because of these competing effects, previous experiments s...

In single-molecule mass spectrometry, the mass of each ion is measured individually; making it suitable for the analysis of very large, heterogeneous objects that cannot be analyzed by conventional means. A range of single-molecule mass spectrometry techniques has been developed, including time-of-flight with cryogenic detectors, a quadrupole ion trap with optical detection, single-molecule Fou...
متن کاملCombined optical trapping and single-molecule fluorescence
BACKGROUND Two of the mainstay techniques in single-molecule research are optical trapping and single-molecule fluorescence. Previous attempts to combine these techniques in a single experiment - and on a single macromolecule of interest - have met with little success, because the light intensity within an optical trap is more than ten orders of magnitude greater than the light emitted by a sin...
متن کاملSingle-molecule fluorescence spectroscopy and imaging
Observability of individual fluorescent molecules under a microscope has many applications in biology.1, 2 In particular, by grafting fluorophores to biomolecules, they can be tracked in real time in live cells for monitoring of their conformational changes and interactions with other molecules. In other words, singlemolecule imaging allows scientists to read the textbook of life pretty much as...
